Mechanisms of copper-binding proteins in (breast) cancer

Copper (Cu) is an essential metal ion acting as a reactive cofactor in many key human enzymes. Since free Cu is toxic due to its redox activity that can damage biomolecules and generate reactive oxygen species, human cells contain an elaborate network of Cu-transport proteins that facilitate specific and timely Cu delivery to target Cu-dependent proteins. The Cu chaperone Atox1 moves Cu(I) in the cytoplasm from the Cu importer Ctr1 to ATPases ATP7A and ATP7B in the Golgi network for subsequent loading of Cu-dependent enzymes in the secretory pathway. Since many proteins depend on Cu for functionality, it is not surprising that cancer cells have a high demand for Cu. Using in vitro biochemical/biophysical experiments combined with cell culture studies, we have found that Atox1 plays a promoting role in cancer cell migration, and we have elucidated the underlying mechanism. We have also discovered that another cancer-promoting protein, Memo1 (mediator of ERBB2-driven cell motility 1), is in fact Cu(I) binding and interacts with Atox1 in a Cu-dependent manner. These results, together with their biological significance, will be discussed in the seminar.
